    Fra le cause di collasso di pendii e rilevati in terra soggetti a filtrazione vi è l’erosione interna di particelle, fenomeno evolutivo che può manifestarsi inizialmente in modo diffuso, per poi localizzarsi in canali di flusso, oppure innescarsi lungo discontinuità preesistenti. Questo contributo presenta lo sviluppo di un’attrezzatura di laboratorio per l’osservazione e la misura del fenomeno di erosione interna e formazione di canali di flusso alla piccola e media scala. Consente di analizzare l’influenza del percorso di flusso e di una pressione di confinamento, di condurre prove in regime di filtrazione confinata o non confinata, e di ricostruire modelli fisici in scala ridotta di pendii e rilevati, per strati di qualunque inclinazione

    Efficient isolation on Vero.DogSLAMtag cells and full genome characterization of Dolphin Morbillivirus (DMV) by next generation sequencing

    The Dolphin Morbillivirus (DMV) genome from the frst Mediterranean epidemic (1990-\u201992) is the only cetacean Morbillivirus that has been completely sequenced. Here, we report the frst application of next generation sequencing (NGS) to morbillivirus infection of aquatic mammals. A viral isolate, representative of the 2006-\u201908 Mediterranean epidemic (DMV_IZSPLV_2008), efciently grew on Vero.DogSLAMtag cells and was submitted to whole genome characterization by NGS. The fnal genome length was 15,673 nucleotides, covering 99.82% of the DMV reference genome. Comparison of DMV_IZSPLV_2008 and 1990-\u201992 DMV strain sequences revealed 157 nucleotide mutations and 47 amino acid changes. The sequence similarity was 98.7% at the full genome level. Whole-genome phylogeny suggested that the DMV strain circulating during the 2006-\u201908 epidemics emerged from the 1990-\u201992 DMV strain. Viral isolation is considered the \u201cgold standard\u201d for morbillivirus diagnostics but efcient propagation of infectious virus is difcult to achieve. The successful cell replication of this strain allowed performing NGS directly from the viral RNA, without prior PCR amplifcation. We therefore provide to the scientifc community a second DMV genome, representative of another major outbreak. Interestingly, genome comparison revealed that the neglected L gene encompasses 74% of the genetic diversity and might serve as \u201chypervariable\u201d target for strain characterization

    Detecting dynamic domains and local fluctuations in complex molecular systems via timelapse neighbors shuffling

    Many complex molecular systems owe their properties to local dynamic rearrangements or fluctuations that, despite the rise of machine learning (ML) and sophisticated structural descriptors, remain often difficult to detect. Here we show an ML framework based on a new descriptor, named Local Environments and Neighbors Shuffling (LENS), which allows identifying dynamic domains and detecting local fluctuations in a variety of systems via tracking how much the surrounding of each molecular unit changes over time in terms of neighbor individuals. Statistical analysis of the LENS time-series data allows to blindly detect different dynamic domains within various types of molecular systems with, e.g., liquid-like, solid-like, or diverse dynamics, and to track local fluctuations emerging within them in an efficient way. The approach is found robust, versatile, and, given the abstract definition of the LENS descriptor, capable of shedding light on the dynamic complexity of a variety of (not necessarily molecular) systems

    TimeSOAP: Tracking high-dimensional fluctuations in complex molecular systems via time variations of SOAP spectra

    Many molecular systems and physical phenomena are controlled by local fluctuations and microscopic dynamical rearrangements of the constitutive interacting units that are often difficult to detect. This is the case, for example, of phase transitions, phase equilibria, nucleation events, and defect propagation, to mention a few. A detailed comprehension of local atomic environments and of their dynamic rearrangements is essential to understand such phenomena and also to draw structure-property relationships useful to unveil how to control complex molecular systems. Considerable progress in the development of advanced structural descriptors [e.g., Smooth Overlap of Atomic Position (SOAP), etc.] has certainly enhanced the representation of atomic-scale simulations data. However, despite such efforts, local dynamic environment rearrangements still remain difficult to elucidate. Here, exploiting the structurally rich description of atomic environments of SOAP and building on the concept of time-dependent local variations, we developed a SOAP-based descriptor, TimeSOAP (Ď„SOAP), which essentially tracks time variations in local SOAP environments surrounding each molecule (i.e., each SOAP center) along ensemble trajectories. We demonstrate how analysis of the time-series Ď„SOAP data and of their time derivatives allows us to detect dynamic domains and track instantaneous changes of local atomic arrangements (i.e., local fluctuations) in a variety of molecular systems. The approach is simple and general, and we expect that it will help shed light on a variety of complex dynamical phenomena

    Moderately Escalated Hypofractionated (Chemo) Radiotherapy Delivered with Helical Intensity-Modulated Technique in Stage III Unresectable Non-Small Cell Lung Cancer

    Purpose: To assess clinical outcomes and toxicities in patients with stage III unresectable non-small cell lung cancer (NSCLC) treated with a moderately escalated hypofractionated radiotherapy delivered with Helical Intensity-Modulated Technique in combination with sequential or concurrent chemotherapy. Materials and Methods: Sixty-one consecutive patients considered non-progressive after two cycles of induction chemotherapy were treated with a moderately escalated hypofractionated radiation course of 30 daily fractions of 2.25–2.28 Gy each administered in 6 weeks up to a total dose of 67.5–68.4 Gy (range, 64.5–71.3 Gy). Thirty-two received sequential RT after two more cycles (total = 4 cycles) of chemotherapy, while 29 were treated with concurrent chemo-radiation. The target was considered the gross tumor volume and the clinically proven nodal regions, without elective nodal irradiation. Results: With a median follow up of 27 months (range 6–40), 1-year and 2-year OS rate for all patients was 77 and 53%, respectively, with a median survival duration of 18.6 months in the sequential group and 24.1 months in the concomitant group. No Grade ≥4 acute and late toxicity was reported. Acute Grade 3 treatment-related pneumonitis was detected in 10% of patients. Two patients, both receiving the concurrent schedule, developed a Grade 3 acute esophagitis. The overall incidence of late Grade 3 lung toxicity was 5%. No patients experienced a Grade 3 late esophageal toxicity. Conclusion: A moderately hypofractionated radiation course delivered with a Helical Intensity-Modulated Technique is a feasible treatment option for patients with unresectable locally advanced NSCLC receiving chemotherapy (sequentially or concurrently). Hypofractionated radiotherapy with a dedicated technique allows safely dose escalation, minimizing the effect of tumor repopulation that may occur with prolonged treatment time

    Mappatura della letteratura sulle soluzioni digitali e tecnologiche nell'assistenza infermieristica: un protocollo di scoping review

       INTRODUCTION: Digital and technological solutions (DTS) might have an impact on people’s personal and professional lives. These types of solutions, according to studies, have the potential to revolutionize and improve the quality and long-term sustainability of healthcare activities, with nurses playing a significant role. Although DTS appears to be intimately linked to the future of nursing, technology must be utilized as an active rather than passive tool. Nonetheless, understanding DTS appears to be difficult, and a scoping study can provide a thorough overview of such a complicated topic. As a result, the scoping study on this topic will map all of the important aspects of DTS and synthesize studies on the nursing workforce, as well as analyze and clarify knowledge gaps and aid future research and development. This article presents the study protocol.  METHODS: The Joanna Briggs Institute (JBI) scoping review methodology will be used for the proposed scoping review. It will include both quantitative and qualitative scientific research as well as grey literature on DTS in nursing. Only English-language works will be considered for inclusion. Two independent reviewers will take part in an iterative process of evaluating literature, choosing papers, and extracting data. Disagreements among reviewers will be resolved through debate until a consensus is reached or through consultation with the study team if necessary. Results will be presented using descriptive statistics, diagrammatic or tabular displayed information, and narrative summaries, as specified in the JBI guidelines.  DISCUSSION: This scoping review protocol explained why it is important to describe the literature on embracing DTS in the nursing field, how to approach the research process, and what the study's key implications will be. The protocol itself may be helpful to increase transparency in the research process, attract interested researchers to work with the group that developed the protocol and offer a practical methodological benchmark for researchers interested in performing scoping reviews by serving as an example of a scoping review protocol.  KEYWORDS: Digital solutions; Technological solutions; Nursing; Scoping review; Study protocol; Framework    INTRODUZIONE: Le soluzioni digitali e tecnologiche potrebbero avere un impatto sulla vita personale e professionale della popolazione. Questi tipi di soluzioni, secondo gli studi, hanno il potenziale per rivoluzionare e migliorare la qualitĂ  e la sostenibilitĂ  a lungo termine delle attivitĂ  di salute, deve gli infermieri svolgono un ruolo significativo di innovatori, facilitatori e gestori dei processi. Sebbene le soluzioni digitali e tecnologiche sembrino essere intimamente legate al futuro dell’assistenza infermieristica, la tecnologia deve essere utilizzata come strumento attivo, piuttosto che passivo. Tuttavia, la comprensione del fenomeno sembra essere difficile e una scoping review può fornire una panoramica completa di un argomento di tale complessitĂ . Di conseguenza, la presente scoping review mapperĂ  tutti gli aspetti salienti legati alle soluzioni digitali e tecnologiche e sintetizzerĂ  gli studi sul coinvolgimento della professione infermieristica, nonchĂ© analizzerĂ  e chiarirĂ  le lacune di conoscenza presenti e aiuterĂ  la ricerca e i progetti futuri. Questo articolo presenta il protocollo di studio di scoping review per raggiungere l’obiettivo di mappatura della letteratura.  METODI: Il protocollo utilizza la metodologia di scoping review del Joanna Briggs Institute (JBI). ComprenderĂ  sia la ricerca scientifica quantitativa che qualitativa, nonchĂ© la letteratura grigia sulle soluzioni digitali e tecnologiche nell’assistenza infermieristica. Solo i lavori in lingua inglese saranno presi in considerazione per l’inclusione. Due revisori indipendenti prenderanno parte a un processo iterativo di valutazione della letteratura, scelta dei documenti ed estrazione dei dati. I disaccordi tra i revisori saranno risolti attraverso il dibattito fino al raggiungimento di un consenso o attraverso la consultazione con il gruppo di studio, ove necessario. I risultati saranno presentati utilizzando statistiche descrittive, le informazioni presentate in forma grafica, tabulare o a mezzo narrativo, come specificato nelle linee guida JBI.  DISCUSSIONE: Questo protocollo di scoping review intende definire perchĂ© è importante descrivere la letteratura sull’adozione delle soluzioni digitali e tecnologiche in ambito infermieristico, come affrontare il processo di ricerca sottostante alla revisione e quali saranno le implicazioni chiave dello studio. Il protocollo stesso può essere utile per aumentare la trasparenza nel processo di ricerca, ingaggiare i ricercatori interessati a lavorare con il gruppo che ha sviluppato il protocollo (research engagement) e offrire un punto di riferimento metodologico e pratico per i ricercatori interessati a svolgere revisioni nel medesimo ambito.  PAROLE CHIAVE: Soluzioni digitali; Soluzioni tecnologiche; Infermieristica; Revisione della letteratura; Protocollo di studi

    <p>Abstract</p> <p>Background</p> <p>Oncogenetic counselling is seldom followed through, even when individuals are eligible according to the test criteria. The basic variables which influence the decision to undergo the genetic counselling process are: risk perception, expected benefit or limitations of genetic testing, general psychological distress or cancer-specific distress, lack of trust in one's emotional reactions when faced with negative events, expected level of family support and communications within the family. The aim of this study was to describe the psychosocial variables of an Italian sample that forgoes genetic counselling.</p> <p>Methods</p> <p>From May 2002 to December 2006 a psychological questionnaire was sent out to one hundred and six subjects, who freely requested a first genetic informative consultation, and never asked to have a second visit and the family tree drawn up in order to inquire about their eligibility for genetic testing. Statistical analysis was performed by Pearson chi-square test, t-test and Spearman RHO coefficient.</p> <p>Results</p> <p>The survey presents a lack of emotional cohesion and structured roles and rules within the family system and a positive correlation between the number of children, anxiety and risk perception. The main reasons for giving up on counselling were a sense that testing was a waste of time and the inability to emotionally handle the negative consequences of the test outcome. The subjects who maintained that test and an early diagnosis were a "waste of time" experienced more anxiety.</p> <p>Conclusion</p> <p>The study revealed the importance to ac knowledging the whole persona and their family system as well as provide information highlighting usefulness of early diagnosis.</p

    Automatic Middle-Out Optimisation of Coarse-Grained Lipid Force Fields

    Automatic data-driven approaches are increasingly used to develop accurate molecular models. But the parameters of such automatically-optimised models are typically untransferable. Using a multi-reference approach in combination with an automatic optimisation engine (SwarmCGM), here we show that it is possible to optimise coarse-grained (CG) lipid models that are also transferable, generating optimised lipid force fields. The parameters of the CG lipid models are iteratively and simultaneously optimised against higher-resolution simulations (bottom-up) and experimental data (top-down references). Including different types of lipid bilayers in the training set guarantees the transferability of the optimised force field parameters. Tested against state-of-the-art CG lipid force fields, we demonstrate that SwarmCGM can systematically improve their parameters, enhancing the agreement with the experiments even for lipid types not included in the training set. The approach is general and can be used to improve existing CG lipid force fields, as well as to develop new custom ones.Comment: Paper (Pages 1-16) + Supporting Information (Pages 17-40
